Effect of CCI©þon ATPase Activity of Mitochondrial Fraction from Rat Liver

Abstract
Since the introduction of Skou¢¥s hypothesis that an Na-K activated adenosine triphosphatase(ATPase) might be involved in the active transport of cation, characterization of this enzyme activity has been widely studied.
The author investigated the activities of Mg ATPase and Na-K ATPase of the mitochondrial fraction of the rat liver by the admininistration of CCI, with or without Panax Ginseng extract in vitro or vivo. The results obtained are summarized as follows:
1. The presence of magnesium ions was necessary for the activity of Na-K ATPase. The Na-K ATPase activity was increased in proportion to the protein content in the mitochondrial fraction.
2. Ouabain, one of the cardiac glycosides, inhibited the activity of Na-K ATPase only, but not the activity of Mg ATPase.
3. The activities of Mg ATPase and Na-K ATPase were both inhibited markedly by CC], in vitro.
4. Administration of CC], to the rat resulted in marked depression of both Mg and Na-K ATPase activities of liver mitochondria) fraction. Single injection of CC], in the doses of 0.5ml/100gm body wt. showed more depression than multiple injections of 0.2m1/100gm body wt. twice a week for two weeks.
5. Inhibition of the Na-K ATPase activity induced by CCI, was lessened by the addition of Panax Ginseng extract in vitro.
In vivo, the Na-K ATPase activity depressd by CCI, was activated by giving Panax Ginseng extract to the rat.
